摘要:目的:本课题旨在研究葡萄籽原花青素提取物对DDP所致大鼠生殖毒性的保护作用,从而为保护机体正常组织且不影响DDP抗肿瘤效果提供一种临床上更为有效的辅助用药。

   方法:成熟雄性SD大鼠50只,随机分为5组。分别为对照组、DDP处理组、DDP+GSPE(200mg/kg)组、DDP+GSPE(400mg/kg)组、GSPE(400mg/kg)组。灌服GSPE后第9天DDP组和DDP+GSPE组一次性腹腔注射DDP(7.0mg/kg),对照组注射等量生理盐水。实验第11天晚上断食,第12天进行动物宰杀。取睾丸及附睾,迅速用液氮冷冻,于-80℃保存。观察各组大鼠组织切片、精子畸形率、谷胱甘肽过氧化物酶(GSH-PX)、超氧化物歧化酶(SOD)、丙二醛指标。

   结果:葡萄籽原花青素提取物可以控制精子畸变;改善DDP引起的睾丸组织SOD和GSH-PX活性下降,降低组织中丙二醛的含量。

   结论:GSPE经口给予能明显预防DDP所致的生殖毒性。其机制可能是GSPE使DDP在体内产生的自由基减少,脂质过氧化程度减轻,提高机体抗氧化能力。

Abstract:Objective: This project is to study the protective effect of grape seed proanthocyanidin extract on cisplatin-induced reproductive toxicity, so as to protect the body from normal tissue without affecting the antitumor effect of cisplatin to provide a clinically more effective adjuvant. 

   Methods: Mature male SD rats were 50, were randomly divided into 5 groups. For the control group, DDP treatment group, DDP + GSPE (200g/kg) group,DDP + GSPE (400g/kg) group, GSPE (400g/kg) group. Gavage the GSPE After nine days of DDP group and the DDP + GSPE, it was a single intraperitoneal injection of the DDP (7.0mg/kg), control group was injected with normal saline. Experiment 11 days of fasting, 12 days for animals slaughtered. Testis and epididymis, and quickly frozen in liquid nitrogen and stored at -80 ℃. Observe the rats in each group tissue sections, sperm deformity rate, glutathione peroxidase (GSH-PX), superoxide dismutase (SOD), malondialdehyde indicators.

   Results: grape seed proanthocyanidin extract can control the sperm aberration; to reduce DDP-induced organ coefficient levels; improve the DDP-induced testicular tissue SOD and GSH-PX activity decreased, reducing the content of malondialdehyde

   Conclusion: GSPE given orally significantly to the prevention of DDP-induced reproductive toxicity. The mechanism may be to The GSPE make DDP to reduce the free radicals generated in vivo, reduce the degree of lipid peroxidation, increase the antioxidant capacity.
